【中文摘要】 利用有機溶劑萃取法提取樹舌靈芝MP-01發(fā)酵液中靈芝酸,并用分光光度法測定提取液中靈芝酸的含量,確定了分光光度法測定的最佳比色條件為:比色波長為520 nm,5%香草醛-冰醋酸和高氯酸用量分別為0.2 mL和1.0 mL(在反應系統(tǒng)中濃度分別為1.0%和83%),顯色溫度70℃,顯色時間為25 min,在此條件下測得醇溶物和醇沉物提取液中靈芝酸含量分別為0.022 mg/g和0.049 mg/g。
【英文摘要】 Ganoderic acid produced during the growth of Ganoderma MP-01 in submerged culture was extracted using ethanol and chloroform,and its concentration was determined spectrophotometrically at 520 nm using oleonolic acid as the standard.Optimal assay conditions were: vanillin concentration 1%,perchloric acid concentration 83%,reaction temperature 70 ℃;reaction time 25 min.Ganoderic acid concentrations in SIE(substance insoluble in ethanol) and SSE(substance soluble in ethanol) fractions were 0.049 mg/g and 0.022 mg/g,respectively.
