【中文摘要】 為驗(yàn)證新型SRAP分子標(biāo)記技術(shù)用于靈芝(Ganoderma lucidum)原生質(zhì)體融合子鑒定的可行性,通過(guò)親本菌株赤芝05、06對(duì)153個(gè)SRAP引物組合進(jìn)行篩選,應(yīng)用經(jīng)過(guò)初篩和復(fù)篩得到的4個(gè)清晰穩(wěn)定、且含有兩個(gè)親本各自特異擴(kuò)增條帶的SRAP引物組合,對(duì)2個(gè)親本菌株、2個(gè)只與親本05有拮抗的融合子、2個(gè)只與親本06有拮抗的融合子、30個(gè)與雙親均有拮抗的融合子進(jìn)行SRAP擴(kuò)增。結(jié)果表明,2個(gè)只與親本05有拮抗的融合子僅擴(kuò)增出06的特異條帶,2個(gè)只與親本06有拮抗的融合子僅擴(kuò)增出05的特異條帶,30個(gè)與雙親均有拮抗的融合子中,有7個(gè)融合子同時(shí)擴(kuò)增出05和06的特異條帶,證明此7個(gè)融合子應(yīng)是05和06融合產(chǎn)生的菌株,也證明SRAP標(biāo)記可以用于靈芝原生質(zhì)體融合子的鑒定。
【英文摘要】 SRAP makers were used to identify protoplast fusion hybrids of Ganoderma lucidum.A total of 153 SRAP primer combinations were selected using G.lucidum parental strains 05 and 06.Four primer pairs generated clear,stable and specific bands for each of the parental strains,and were used to confirm protoplast fusion hybrids.A total of 181 strains were isolated from regenerated protoplasts after fusion treatment of equal numbers of protoplasts prepared from each of the two parental strains.These strains were divided into three groups based on their antagonistic effect with the two parental strains: 35 isolates exhibited antagonism with parental strain 05(Group A),116 isolates with parental strain 06(Group B),and 30 isolates with both parental strains(Group C).SRAP amplification was then carried out with the two parental strains,with two isolates each from A and B groups,and with all the Group C isolates using the four selected SRAP primer pairs.Bands specific only for parental strain 06 wer...更多e observed with Group A isolates,whereas Group B isolates generated bands specific only for parental strain 05.However,bands specific for both parental strains were observed with seven Group C isolates,thereby confirming these isolates to be true protoplast fusion hybrids. 還原
