【中文摘要】 為建立雙孢蘑菇同核原生質(zhì)體雜交育種體系,本文對(duì)同核原生質(zhì)體親合性反應(yīng)及雜交異核體F_1代的農(nóng)藝性狀和F_2代的分離與變異規(guī)律進(jìn)行了研究。結(jié)果表明:雙孢蘑菇同核原生質(zhì)體的雜交是一個(gè)較復(fù)雜的性親合與核遷移過程,其雜交率較低。在按氣生型×氣生型、氣生型×匍匐型、匍匐型×匍匐型組合的520個(gè)雜交配對(duì)中,共獲得113個(gè)雜交異核體,平均雜交率為22%。本研究同時(shí)建立了以菌落形態(tài)、菌絲生長(zhǎng)速度、羧甲基纖維素酶相對(duì)活性以及酯酶同功酶電泳為手段的雜交異核體鑒定體系。
【英文摘要】 Compatible reaction of homokaryotic protoplasts, agronomic characters of heterokaryotic hybrid F1 generations, separation and variation of F2 generations were studied in this paper to establish a cross breeding system of homokaryotic protoplast in Agaricus bisporus. Results showed that cross between homokaryotic protoplast in Agaricus bisporus was a complicated procedure of sex compatibility and nuclear migration and its hybridization percentage was not high. 113 heterokaryotic hybrids were obtained from 520 combinations pairing with fluffy type x fluffy type, fluffy type x appressed type, appressed type x appressed type and average hybridization percentage was 22%. Meanwhile, identification system for heterokaryotic hybrid was also established by means of colonial morphology, mycelial growth rate, relative activity of Cx enzyme and esterase isozyme elecrophoresis.
