【摘要】 液體培養(yǎng)蛹蟲草(Cordyceps militaris)魯山株C0511,以菌絲體產(chǎn)量為指標,對培養(yǎng)時間、接種量、溫度和培養(yǎng)基起始pH進行單因素試驗,結(jié)果表明,最佳培養(yǎng)時間為96h,接種量為10%,適宜溫度為22~26℃,pH為5.8。正交試驗結(jié)果表明,影響蛹蟲草菌絲液體培養(yǎng)的主次因素依次為培養(yǎng)溫度、培養(yǎng)時間、接種量和pH。魯山菌株C0511液體菌種按10%的接種量接入起始pH為7.0的液體培養(yǎng)基中,25℃培養(yǎng)96h菌絲產(chǎn)量為30.13g/L。
【Abstract】 Monofactorial testing revealed that mycelial yields of Cordyceps militaris (strain Lu Shan C0511) grown in submerged culture were maximal under the following conditions:culture temperature 2226 ℃,incubation time 96 h,inoculum size 10% (v/v),medium pH 5.8. Application of orthogonal testing revealed that mycelial yields were influenced predominantly by culture temperature,followed by incubation time,inoculum size,and the initial pH of the medium. When 10% (v/v) liquid spawn of strain Lu Shan C0511 was inoculated into medium adjusted to pH 7.0 and incubated at 25 ℃ for 96 h,the mycelium yield was 30.13 g/L.
