應用iTRAQ結合2D LC-MS/MS技術分析草菇同核和異核菌絲蛋白質組

【摘要】 首次利用同位素標記相對和絕對定量(isobaric tags for relative and absolute quantification,iTRAQ)技術結合二維液相色譜串聯(lián)質譜對草菇(Volvariella volvacea)同核和異核菌絲蛋白質組進行研究。采用QExactive質譜鑒定并經(jīng)MASCOT軟件搜庫,對所有鑒定蛋白進行主成分分析(principal component analysis,PCA)、層次聚類(hierarchical clustering)分析和GO(gene ontology)注釋,并進行生物信息學分析。結果共獲得2335個不同肽段,鑒定到1039個蛋白,其中1030個蛋白具有定量信息。在同核菌絲中顯著上調蛋白83個,下調蛋白97個,表明iTRAQ標記技術結合二維液相色譜串聯(lián)質譜可有效地分離和鑒定草菇菌絲蛋白質組。該研究結果為更深入全面地研究草菇菌絲蛋白質組學奠定基礎。
【Abstract】 Protein expression in homokaryotic and heterokaryotic mycelia of Volvariella volvacea has been investigated using isobaric tags for relative and absolute quantification(iTRAQ)-coupled 2D LC-MS/MS(two-dimensional liquid chromatography tandem mass spectrometry).A total of 1039 proteins,including 2335 unique peptides,were identified using Q Exactive mass spectrometry and the MASCOT search engine,and 1030 of these had a quantitative dimension.Of the quantitatively different proteins,83 were signific...
