【摘要】 采用組織分離法從市售牛肝菌子實體分離得到兩株性狀穩(wěn)定的分離株Lq4-2和Be2425,利用ITS序列分析并結(jié)合分離子實體的形態(tài)特征對兩分離株進行鑒定,并對其基礎(chǔ)生物學特性進行了研究。分離株Lq4-2鑒定為牛肝菌科(Boletaceae)成員,分離株Be2425鑒定為廣義美味牛肝菌(Boletus eduliss.l.);兩分離株能在PDA培養(yǎng)基和MRD合成培養(yǎng)基上良好生長;菌絲生長的適宜溫度為2528℃,適宜pH為56,最佳氮源為酒石酸銨;Lq4-2的最佳碳源為葡萄糖,而Be2425的最佳碳源為果糖和葡萄糖。
【Abstract】 Two bolete strains,Lq4-2 and Be2425,were isolated using standard tissue culture methodology from fruit bodies purchased at two local markets,and assigned to the Boletaceae and Boletus edulis sensu lato,respectively based on internal transcribed spacer(ITS1-5.8S-ITS2) sequence analysis.Both strains exhibited highest mycelial growth rates and growth vigor on potato dextrose agar,although several other media also supported fungal growth.Both strains grew optimally between 2528 ℃,between pH 56,and with ammonium tartrate as the N source.Among several carbon sources tested,glucose supported the highest Lq4-2 growth rates,and either fructose or glucose in the case of Be2425.
