【摘要】 根據(jù)草菇(Volvariella volvacea)S-腺苷-L-高半胱氨酸水解酶基因(sahh)全長序列設(shè)計(jì)兩個嵌套引物,與Takara LA PCR in vitro cloning試劑盒的接頭引物結(jié)合進(jìn)行巢式PCR擴(kuò)增,得到sahh上游調(diào)控序列,通過序列分析發(fā)現(xiàn),該序列含有1個TATA盒和4個CAAT盒。應(yīng)用MatInspector程序分析sahh上游調(diào)控序列,發(fā)現(xiàn)該區(qū)域存在1個耐鹽因子,1個熱激因子,2個DRE元件和2個ABA應(yīng)答元件。
【Abstract】 An upstream regulatory sequence of a S-adenosyl-L-homocysteine hydrolase gene(sahh) from Volvariella volvacea was amplified by nested PCR using two nested primers designed according to the full-length sequence of sahh and which were connected to the primer linker of the Takara LA PCR in vitro cloning box.The upstream sequence contained one TATA box and four CAAT boxes.MatInspector program analysis revealed one salt resistance factor,one heat shock factor,two drought-response binding factors(DRE)...
